Whereas the 2-substituted-thiomorpholine pharmacophore is currently considered of high importance in medicinal chemistry, so far only few general methods have been developed for its preparation in an enantiomerically pure form. This review covers the available methods affording 2-substituted-thiomorpholines in a racemic form together with the recent progresses accomplished in the asymmetric or solid phase synthesis domain.
