ABSTRACT
 
The droplet vitrification method, a one-step freezing technique, has been used as a biotechnological strategy for the long-term preservation of ‘Gökova Sesame’, which is known as the highest quality sesame in Turkey. Gökova sesame acts as an antioxidant as it is rich in secondary metabolites and phytate compounds. Antioxidants reduce the effects of free oxygen radicals and are also effective against cancer. There is no study or strategy available in the literature for the long-term preservation of ‘Gökova Sesame’. This local sesame variety is grown in a small area, making the preservation of its germplasm particularly important. The cryopreservation method is a technique that provides long-term preservation by keeping it in liquid nitrogen at -196 °C. The aim of this study is to develop an effective procedure for the long-term preservation of the local sesame cultivar through vitrification and one-step freezing techniques. In this method, after cold hardening and sucrose preculture steps, in vitro grown Gökova sesame meristems were placed on aluminum strips and held in 4 μl PVS2 solution for different time periods, and then transferred directly to liquid nitrogen. The recovery rate obtained after the thawing process was 40% for the pre-study. This study could be considered as a significant step in the preservation of Gökova Sesame and would be beneficial for future research.
