ABSTRACT The epitopes of antibodies generated in hemophiliacs using synthetic peptides were studied. In addition, neutralization of the antibodies using the epitope peptides was tried. Peptides strongly combined with the antibodies using substitution peptides were determined so that the antibodies could be neutralized by low-concentration peptides. We have determined the location of epitopes on the factor IX for three Japanese hemophilia B inhibitor antibodies and the factor VIII for nine Japanese hemophilia A inhibitor antibodies using synthetic peptides. Additionally, changes of antigenicity using the systematic substitution of different amino acids at each residue of the epitope peptides and neutralization rate of factor IX inhibitors by a lower concentration of substitutional synthetic peptides conjugated with bovine serum albumin are reported.
